The Potential of Cinnamon Extract (Cinnamomum burmanii) as Anti-insomnia Medication through Hypothalamus Pituitary Adrenal Axis Improvement in Rats.

This study aimed to explore the efficacy of cinnamon extract as an anti-insomnia medication in experimental animals by evaluating the levels of hormones and neurotransmitters related to insomnia. A total of 30 male Wistar rats were divided into six groups. Induction of insomnia in animal models was done by administration of p-chloro-phenylalanine (PCPA) compounds. Estazolam was administrated to the positive control group. Cinnamon extract administration was divided into 3 doses, namely: 25 mg/kg BW, 50 mg/kg BW and 100 mg/kg BW. Evaluation of the organ coefficient was conducted to evaluate drug toxicity to the organs. The enzyme-linked-immunoassay method assessed hormones and neurotransmitters in the serum and hypothalamus related to insomnia. There was a decrease in the adrenal coefficient in the cinnamon extract group compared to the PCPA group (0.011+0.001, P<0.05). In addition, there was a decrease in the corticotropin-releasing hormone, adrenocorticotropin hormone, and corticosterone levels in the serum of animals who received cinnamon extract. Our study found a dose of cinnamon extract of 50 mg/kg BW was the best dose to balance neurotransmitter levels in insomniac rats. The cinnamon extract increased serotonin and melatonin levels and decreased norepinephrine levels in the insomnia-induced group. Cinnamon extract has potential as an anti-insomnia medication through hypothalamus-pituitaryadrenal axis improvement and brain neurotransmitter regulation in an animal model of insomnia.

Cinnamomum cassia extract promotes thermogenesis during exposure to cold via activation of brown adipose tissue

Ethnopharmacological relevanceCinnamomum cassia (L.) J.Presl (Lauraceae), a widely used traditional Chinese medicine, is well known to exert hot property. It is recorded as dispelling cold drug in ancient Chinese monographs, such as Synopsis of golden chamber published in Han dynasty. According to Chinese Pharmacopoeia (2015), Cinnamomum cassia (L.) J.Presl (Cinnamon) has the functions of dispersing cold, relieving pain, warming meridians and promoting blood circulation. Aim of the studyThe aim of this study is to evaluate the effect of Cinnamon extract (CE) on cold endurance and the mechanism of thermogenesis activity. Materials and methodsThe improving effect of hypothermia were evaluated with body temperature by infrared camera and multi-thermo thermometer. In vivo, the thermogenic effect was observed with energy metabolism and substrate utilization. The activation of brown adipose tissue (BAT) was evaluated with the histomorphology and expression of thermogenic protein. In vitro, the uncoupling effect on mitochondrial was evaluated with Seahorse and fluorescent staining. The mechanism of thermogenesis was explored in brown adipocyte. ResultsThe body temperature and energy expenditure were significantly increased by CE administration in cold environment. In morphology, lipid droplets were reduced and the number of mitochondrial was increased. CE significantly increased the non-shivering thermogenesis via upregulating the expression of thermogenic protein. In vitro, the uncoupling effect was obviously along with the decreased mitochondrial membrane potential and ATP production. It was confirmed that the thermogenesis effect was induced via lipolysis and energy metabolism. In addition, CE also alleviated myocardium injury in the morphology in cold environment. Moreover, the major constituent was identified as (1) coumarin, (2) cinnamic acid, (3) cinnamaldehyde and (4) 2-methoxy cinnamaldehyde. ConclusionsThe mechanism of improving cold tolerance was related to lipolysis and activation of BAT. Meanwhile, we provided a kind of potential prevention methods for cold injury.

EFFECT OF SODIUM VALPROATE ON THE STRUCTURE OF THE RENAL CORTEX OF ADULT MALE ALBINO RAT AND THE ROLE OF CINNAMON

Background: Valproic Acid (VPA) is one of the most widely prescribed antiepileptic drugs and is regarded as a first choice for most forms of seizures. Although valproic acid has a wide therapeutic window, yet it is associated with many adverse effects. Objectives: Evaluation of the effect of sodium valproate on renal cortex structure of adult male albino rats, and the role of cinnamon. Materials and Methods: Twenty four adult male albino rats weighing 160-180 grams were divided into four equal groups: Group I: control group, group II: sodium valproate group (500 ml/kg/day orally), group III: Cinnamon extract group (4ml/rat/day orally) and group IV: sodium valproate and cinnamon extract group. Drugs were given orally once daily for four weeks.      Kidney samples were collected for histological, imunohistochmical and ultrastructural studies. Blood urea nitrogen (BUN) and serum creatinine (sCr) concentration were measured. Morphometric quantitative study for area percentage of collagen fibers and Caspase-3 +ve cells were done. Results: Examination of the renal cortex of sodium valproate ingested group demonstrated atrophied glomerulus with collapsed tuft, widening of bowman's space, degenerated tubule, hemorrhage and cellular infiltration. Moreover, the collagen fibers increased as well as focal loss of the basement membrane and brush border of the tubules.  Caspase-3 +ve cells significantly increased after sodium valproate administration. Rarefaction of the cytoplasm, degeneration of mitochondria, loss of apical microvilli and pyknotic nucleus were observed in the electron microscopically study.      The kidney functions demonstrated significant increase in   concentration of blood urea nitrogen (BUN) and serum creatinine (sCr). In contrast, cinnamon extract administration with sodium valproate ingested group showed some amelioration in the renal cortex structure and function. Conclusion: Sodium valproate aggravated the histological structure of the renal cortex. However, cinnamon extract could ameliorate the changes due to its antioxidants effect.

Hypolipidemic and Hypoglycemic Effect of Cinnamon Extract in High Fat Diet Fed Rats

The purpose of this study was to evaluate the effects of oral administration of cinnamon extract (CE) in rats fed on high fat diet (HFD). Thirty two adult male albino rats were divided into four groups; control group, HFD group, HFD group treated by cinnamon and the last group was treated with cinnamon extract. In HFD group, rats were fed on HFD for two months. In HFD group treated by cinnamon, rats fed on HFD for two months then CE was orally administered for a period of 4 weeks at a dose of 200 mg/kg body weight. In CE treated group, rats were fed on standard diet for two months then orally administered CE for a period of 4 weeks at a dose of 200 mg/kg body weight. The obtained results indicated that the oral administration of cinnamon extract has antihyperlipidemic effect that improving the lipid profile. There were reductions in the levels of serum triglycerides, LDL-C, VLDL-C (P≤ 0.01) and total cholesterol (P<0.05) with an increase of the HDL-C serum level (P≤ 0.01) in HFD fed rats treated by CE. There was a significant decrease in blood glucose level (P≤ 0.01), elevation of serum insulin level (P≤ 0.01) and reduction in liver enzyme ALT and AST activities (P≤ 0.01) with oral administration of CE. In addition, CE can promote glucose transporter isotype-4  (GLUT4) gene expression in adipose tissue of HFD fed rats. From the current study, it was concluded that oral administration of cinnamon extract has both hypolipidemic and hypoglycemic effects in hyperlipidemic rats.

Cinnamon induces browning in subcutaneous adipocytes

Browning is the process of increasing the number of brite cells, which helps to increase energy expenditure and reduce obesity. Consumption of natural and non-toxic herbal extracts that possess the browning effect is an attractive anti-obesity strategy. In this study, we examined the browning effect of cinnamon extract. We found that cinnamon extract (CE) induced typical brown adipocyte multiocular phenotype in 3T3-L1 adipocytes. The treatment also increased brown adipocytes markers and reduced white adipocytes markers in the 3T3-L1 adipocytes. In ex vivo studies, we found that CE increased brown adipocytes markers in the subcutaneous adipocytes isolated from db/db mice and diet-induced obesity (DIO) mice. However, CE did not significantly affect UCP1 expression in the adipocytes isolated from perinephric adipose tissue and epididymal adipose tissue. β3-adernergic receptor (β3-AR) antagonist reduced the CE-enhanced UCP1 expression, suggesting an involvement of the β3-AR activity. Oral administration of CE significantly increased UCP1 expression in the subcutaneous adipose tissue in vivo and reduced the body weight of the DIO mice. Taken together, our data suggest that CE has a browning effect in subcutaneous adipocytes. Our study suggests a natural non-toxic herbal remedy to reduce obesity.

The effect of Cinnamomum zeylanicum bark water extract on memory performance in alloxan-induced diabetic mice.

Cinnamomum zeylanicum (cinnamon) has a wide range of beneficial effects including mild glucose lowering activity. The aim of the present study was to investigate whether cinnamon bark extract has the potential to improve memory performance and glucose profiles in diabetic mice. Memory was assessed by the novel object recognition task in male Balb/c mice. In this method, the difference between exploration time of a familiar object and a novel object was considered as an index of memory performance (recognition index, RI). The water extract was prepared by boiling cinnamon bark for 15 min. Alloxan induced diabetes in animals (serum glucose levels were 322 ± 7.5 mg/dL), and also impaired memory performance (RI= -3.3% ± 3.3) which differed significantly from control animals (RI = 32% ± 6.5). Although treatment with cinnamon only reduced fasting blood glucose level moderately but it improved memory performance remarkably (RI = 25.5% ± 5.6). Oxidative stress following administration of cinnamon extract was lower in diabetic mice. It was concluded that cinnamon water extract could be a useful alternative medicine in diabetic patients’ daily regimen which not only reduces blood glucose levels but also improves memory performance and lipid peroxidation level.

Studies on the biochemical and molecular effects of some natural herbs on experimental-induced breast cancer in Wistar rats

Background: Breast cancer is the most frequent malignant tumor in women, and its metastatic state represents the second cause of mortality. To treat cancer, we must prevent tumor cell proliferation and angiogenesis. The usage of herbal medication for treatment of some diseases as diabetes is increased nowadays for their wide safety margin. Aims and Objectives: Therefore, the current study was designed to examine the protective effect of ginger extract (GE) and cinnamon extract (CE) in controlling the mammary gland incidence in female Wistar rats. Materials and Methods: A total of 75 female virgin Wistar rats were allocated into 5 groups. Control group without any treatment; mammary gland group administered 7, 12-dimethyl-benz[a]anthracene at a dose of 20 mg/kg orally in corn oil. Rats in 3-5 served as tumor groups and received GE for group 3 (0.125% in water), CE for group 4 (100 mg/kg), and a mixture of GE plus CE for group 5. GE and CE were administered 2 weeks before cancer induction and continued for 4 months. Serum levels of breast cancer biomarker (CA125) and oxidative stress biomarkers were measured. Tissues of tumors were examined at molecular and histopathological levels. Results: There was a significant increase in the serum levels of CA125 in untreated mammary gland tumor group when compared with untreated healthy rats. There was a significant decrease in the serum levels of glutathione (GSH) peroxidase, catalase, superoxide dismutase, and GSH reductase in the untreated mammary gland tumor groups compared to control healthy rats. Administration of ginger and CE normalized the decrease in antioxidants levels. Induction of mammary gland tumor upregulated the genes associated with tumor incidences such as GST-P, CYP1A1, CYP1B1, and vascular endothelial growth factor-receptor 1 and administration of both herbal plants normalized these changes. Coadministration of GE and CE-induced additive inhibitory effect on genes upregulated due to tumor incidence. Bax expression was downregulated in tumor group and increased after GE and CE expression either alone or in combination. At histopathological levels, breast cancer group showed adenocarcinoma that is decreased with plants extracts administration when compared with breast cancer and control group. Conclusion: Results suggest that extracts of ginger and cinnamon have anticarcinogenic protective activities and support the hypothesis that these plants help in the prevention of mammary cancer by controlling the expression of carcinogenesis-associated genes.

Cinnamon extract inhibits degranulation and de novo synthesis of inflammatory mediators in mast cells

Mast cells (MC) are main effector cells of allergic and other inflammatory reactions; however, only a few anti-MC agents are available for therapy. It has been reported that cinnamon extract (CE) attenuates allergic symptoms by affecting immune cells; however, its influence on MC was not studied so far. Here, we analyzed the effects of CE on human and rodent MC in vitro and in vivo. Expression of MC-specific proteases was examined in vivo in duodenum of mice following oral administration of CE. Release of mediators and phosphorylation of signaling molecules were analyzed in vitro in human MC isolated from intestinal tissue (hiMC) or RBL-2H3 cells challenged with CE prior to stimulation by FcεRI cross-linking. Following oral treatment with CE, expression of the mast cell proteases MCP6 and MC-CPA was significantly decreased in mice. In hiMC, CE also caused a reduced expression of tryptase. Moreover, in hiMC stimulated by IgE cross-linking, the release of β-hexosaminidase was reduced to about 20% by CE. The de novo synthesis of cysteinyl leukotrienes, TNFα, CXCL8, CCL2, CCL3, and CCL4, was almost completely inhibited by CE. The attenuation of mast cell mediators by CE seems to be related to particular signaling pathways, because we found that activation of the MAP kinases ERK, JNK, and p38 as well as of Akt was strongly reduced by CE. CE decreases expression of mast cell-specific mediators in vitro and in vivo and thus is a new plant-originated candidate for anti-allergic therapy.

CINNAMON EXTRACT REGULATES GENE EXPRESSION OF LIPID AND CARBOHYDRATE METABOLISM IN STREPTOZOTOCIN INDUCED DIABETIC WISTAR RATS

Cinnamon is a herbal medication plant used widely f or many diseases in the Middle East and ancient countries. Cinnamon Extract (CE) contains many active substances that modulate insulin sensitivity and acts as anti-diabetic medication. Diabetes mellitus is a disease characterized by disturbance in carbo hydrate and lipid metabolism. The aim of this study was to examine the effect of Cinnamon Extract (CE, 200 mg kg -1 body weight) on genes expression of lipid and carb ohydrate metabolism in Streptozotocin (STZ) induced diabetes in Wistar rats. Administration of CE to STZ diabetic rats for 2 months normalized TG, cholesterol, VLDL, HDL and glucose levels. CE administration increased FAS and STREB-1c gene expressions that were inhibited in diabetic rats. M oreover, CE inhibited significantly the increase in LPL, HSL and resistin expression. In parallel, CE increa sed the expression of PPECK and GLUT2 and inhibited the decrement in IGF and PK expression. In conclusi on, CE regulates the expression of genes related to lipid and carbohydrate metabolism in a way to contr ol the metabolic disorders occur during diabetes.

Cinnamon extract prevents the insulin resistance induced by a high-fructose diet.

The aim of this study was to determine whether cinnamon extract (CE) would improve the glucose utilization in normal male Wistar rats fed a high-fructose diet (HFD) for three weeks with or without CE added to the drinking water (300 mg/kg/day). In vivo glucose utilization was measured by the euglycemic clamp technique. Further analyses on the possible changes in insulin signaling occurring in skeletal muscle were performed afterwards by Western blotting. At 3 mU/kg/min insulin infusions, the decreased glucose infusion rate (GIR) in HFD-fed rats (60 % of controls, p < 0.01) was improved by CE administration to the same level of controls (normal chow diet) and the improving effect of CE on the GIR of HFD-fed rats was blocked by approximately 50 % by N-monometyl-L-arginine. The same tendency was found during the 30 mU/kg/min insulin infusions. There were no differences in skeletal muscle insulin receptor (IR)-beta, IR substrate (IRS)-1, or phosphatidylinositol (PI) 3-kinase protein content in any groups. However, the muscular insulin-stimulated IR-beta and IRS-1 tyrosine phosphorylation levels and IRS-1 associated with PI 3-kinase in HFD-fed rats were only 70 +/- 9 %, 76 +/- 5 %, and 72 +/- 6 % of controls (p < 0.05), respectively, and these decreases were significantly improved by CE treatment. These results suggest that early CE administration to HFD-fed rats would prevent the development of insulin resistance at least in part by enhancing insulin signaling and possibly via the NO pathway in skeletal muscle.
